چکیده

Antibody Production The cDNA sequence corresponding to amino acids 1-416 of Vha100-1 was amplified by PCR to incorporate BamHI and NotI restriction sites and then cloned into to the pET28a (Invitrogen) vector for protein expression. The primers used were: 5’-CGGAATTCATGGGTTCCCTATTCCGCAGCGAGG-3’ and 5’-CCGCTCGAGGGATTCATCTCCCGATAGCTGGCC-3’. Guinea pig antibodies against this domain were raised by Cocalico Biologicals (Reamstown, PA) using the purified recombinant protein.

I-6: Role of Actin Cytoskeleton during Mouse Sperm Acrosomal Exocytosis

Background: Mammalian sperm must undergo a process termed capacitation to become competent to fertilize an egg. Capacitation renders the sperm competent by priming the cells to undergo a rapid exocytotic event called acrosomal exocytosis that is stimulated by the zona pellucida (ZP) of the egg or progesterone. Over the years, several biochemical events have been associated with the capacitation...

Tilting the Balance between Facilitatory and Inhibitory Functions of Mammalian and Drosophila Complexins Orchestrates Synaptic Vesicle Exocytosis

SNARE-mediated synaptic exocytosis is orchestrated by facilitatory and inhibitory mechanisms. Genetic ablations of Complexins, a family of SNARE-complex-binding proteins, in mice and Drosophila cause apparently opposite effects on neurotransmitter release, leading to contradictory hypotheses of Complexin function. Reconstitution experiments with different fusion assays and Complexins also yield...

A product of the Drosophila stoned locus regulates neurotransmitter release.

The Drosophila stoned locus encodes two novel gene products termed stonedA and stonedB, which possess sequence motifs shared by proteins involved in intracellular vesicle traffic. A specific requirement for stoned in the synaptic vesicle cycle has been suggested by synthetic genetic interactions between stoned and shibire, a gene essential for synaptic vesicle recycling (Petrovich et al., 1993)...

Ca2+ Influx through Distinct Routes Controls Exocytosis and Endocytosis at Drosophila Presynaptic Terminals

Endocytosis of synaptic vesicles follows exocytosis, and both processes require external Ca(2+). However, it is not known whether Ca(2+) influx through one route initiates both processes. At larval Drosophila neuromuscular junctions, we separately measured exocytosis and endocytosis using FM1-43. In a temperature-sensitive Ca(2+) channel mutant, cacophony(TS2), exocytosis induced by high K(+) d...

Essential function of Drosophila Sec6 in apical exocytosis of epithelial photoreceptor cells

Polarized exocytosis plays a major role in development and cell differentiation but the mechanisms that target exocytosis to specific membrane domains in animal cells are still poorly understood. We characterized Drosophila Sec6, a component of the exocyst complex that is believed to tether secretory vesicles to specific plasma membrane sites. sec6 mutations cause cell lethality and disrupt pla...

Flowers for Synaptic Endocytosis

Exocytosis and endocytosis of synaptic vesicles are tightly coordinated to maintain a steady supply of new vesicles during periods of extended neuronal stimulation. Yao et al. (2009) now report that a synaptic vesicle membrane protein named Flower promotes endocytosis at neuromuscular junctions in the fruit fly Drosophila.
